A Canadian roadpic, directed by Bruce McDonald and starring Don McKellar (et mucho al), involving mass hijinx in the surreptitious transport of some heroin from small-town Ontario to New Orleans. It works better as a film than as an Everything node, trust me. Satan has a cameo or three; nearly steals the show.
The highway upon and alongside which many of the events of the aforementioned film take place. Bob Dylan revisited it once, but that was a couple of decades earlier.

Just to add to pingouin's node, Jello Biafra has a very odd cameo as a border patrol guard at the US-Canadian border. Hearing him rant about "America is my home! How can I invite a criminal into my home?" is priceless.
As for Satan, consider the following: at the crossroads of Highway 61 and Highway 49 in Mississippi, Robert Johnson is said to have called up the devil and sold his soul.
Highway 61 runs from Ottawa, Canada, through Minneapolis, St. Louis, Memphis, down to New Orleans. It is the Mississippi River of American highways.
Finally, Dylan did write a song, but its actual title is "Highway 61 Revisited", like the album.
