The specimen is illuminated by light passing through an annular diaphragm. Direct light rays travel a different path than light rays that are reflected or diffracted as they pass through the specimen. Those two sets of rays are combined at the eye.

This type of microscopy shows greater differentiation of internal structures and shows the sheath wall.
